亮菌多糖的研究——Ⅰ.ATM3组分的分离纯化及其性质
STUDIES ON POLYSACCHARIDES OF LIANGJUN I.ISOLATION AND IDENTIFICATION OF ATM3
【摘要】 亮菌系担子菌。分离于腐烂柳木上,经子实体分离后,进行发酵培养。菌丝用热水抽提、乙醇沉淀、去蛋白、逆向流水透析、DEAE-纤维素柱层析两次,得到白色粉末ATM3。ATM3经超离心密度梯度分析,玻璃纤维纸电泳和Sephadex G-200柱层析,证明为单一均匀成分。ATM3的比旋度为[α]D+95.2°,不含氮,分子量145,000。用红外光谱法、1H核磁共振和13C核磁共振法证明其糖苷键为α型。经纸层析及气相层析分析,ATM3含D-葡萄糖,D-半乳糖,D-甘露糖,L-木糖和L-岩藻糖,其摩尔比为0.86;0.30:3.91:1.0:0.85。ATM3进行过碘酸盐氧化及Smith降解,证明其结构中的主要连接键型为α(1→6),并有少量α(1→3)。ATM3动物半体内肿瘤抑制率为81%。动物体内抑制率对S-180为26.6%,对HAC为37.7%。
【Abstract】 ATMS,a polysaccharide component of Armillariella tabeces(Scop.ex Fr)Sing,called Liangjun,was prepared by ethanol precipitation of the hot water extract ofculture mycelia with subsequent purification through DEAE-cellulose column chromatography.Its homogeneity was examined by density gradient centrifugation,etctrophoresis and Sephadex G-200 chromatography.ATM3 is mainly composed ofα-linked D-glc,D-gal,D-man,L-xyl and L-fuc in the molar proportions of 0.86:0.30:3.91:1:0.85.The estimated molecular weight of ATM3 is 145,000.Opticalrotation [α]_D=+95.2°.Periodate oxidation and Smith-degradation showed that α(1→6) linkages predominate over α(1→3)linkages.The in vitro inhibition rate ofATM3 against Ehrlich ascites carcinoma in mice is 81%,against the in vivo inhibitionrates of 26.6% for sarcoma-180 and 37.7% for HAC.
