大麦条纹花叶病毒(新疆株)中多聚腺苷酰核糖核酸含量、侵染活性及多聚腺苷酸链长分布的测定
THE CONTENT,INFECTIVITY AND DISTRIBUTION OF POLY(A) AT THE 3′TERMINUS OF POLYADENYLATED RNA FROM BARLEY STRIPE MOSAIC VIRUS(XINJIANG STRAIN)
【摘要】 1.用Oligo(dT)纤维素亲和层析将新疆大麦条纹花叶病毒(BSMV-XJ)RNA分为poly(A)+(与Oligo(dT)纤维素结合的)RNA和poly(A)-(不与Oligo(dT)纤维素结合的)RNA,其相对含量poly(A)+RNA占75%,poly(A)-RNA为25%。2.用32P标记法测定了BSMV(XJ)RNA中3′端poly A的链长分布,结果poly(A)+RNA带有数个至三十几个腺苷酸残基,而poly(A)-RNA也含十个以下的腺苷酸残基。3.用系统感病寄主大麦进行侵染性试验表明,poly(A)+RNA与poly(A)-RNA对大麦具有同等水平的侵染性。4.文中讨论了Oligo(dT)纤维素的分离效果以及所用测定poly(A)链长分布方法的可靠性。
【Abstract】 By means of oligo(dT)-cellulose affinity chromatography the intact RNA ofBSMV(Xinjiang strain) can be separated into unbound(poly(A)-RNA) and bound(poly(A)~+ RNA) fractions.The ratio of poly(A)~- RNA and poly(A)~+ RNA in thetotal BSMV-RNA is 1:3.We observed that the length distribution of poly(A) at the3′-terminus of poly(A)~+ RNA is from several to more than 30 residues of adenosineand that the poly(A)~- RNA has also a poly(A) tail of less than 10 residues ofadenosine.The term poly(A)~- RNA as used here therefore refers to RNA that doesnot contain sufficient poly(A) to bind to the affinity cellulose.The infectivity of poly(A)~- RNA from BSMV(Xinjiang strain) is similar to thatof poly(A)~+ RNA suggesting that the length 3′-terminal poly(A) is not essential forinfectivity of BSMV-RNA.:~
