测定小鼠脾混合淋巴细胞反应的新方法——SCM测定法

【摘要】 本文利用近年来新发展的胞浆基质结构性(SCM)方法检测了小鼠脾混合淋巴细胞反应。 实验结果指出,组织相容(近交系动物)个体之间 MLR不表现 SCM反应;组织不相容(不同品系之间)个体则表现 SCM 反应。从 CFW,C57BL,C57BL6,ICR,615,BALB/c,F1ICR/615小鼠得到的结果表明,当组织不相容时,RRSCM((PMLR/PC)×100%)=75±5%,CV<20%,与传统的同位素掺入检测MLR方法结果相符。 实验提示,脾淋巴细胞密度、分离条件及试剂的准备对成功地导出SCM反应有很大关系。本文对SCM反应细胞群等也作了探讨。 该方法与传统的 MLR(5~7天)方法相比,具有快速(全过程 4~5 小时)这一明显优点。本文亦讨论了在临床移植配型工作中应用该方法的可能性。

【Abstract】 This paper reports the adaptation of the newly developed structuredness of cytoplasmic matrix (SCM) technique of Cercek (1974) to the detection of mouse spleen MLR.Experimental results indicated that SCM reaction was only detected in MLK of cells from histoincompatible individuals (mice from different strains) but not in those from histocompatible individuals (inbred strains). For the former, a SCM Eesponse Ratio (RRSCM = PMLR/PC×100%) of 75± 5% (GV<20%) was found. These results were compatible with that obtained by the conventional method of MLC determination.In order to achieve successful adaptation of the technique and induction of SCM reaction, the following details were found important: Density of spleen lymphocytes, experimental conditions in the fractionation of cells, preparation of reagent etc. The population of lymphocytes which were SCM responsive and other relevant factors are discussed.Compared with the conventional method of MLC, the method reported here has the virtue of rapidity 4-5h, which render it more advantageous for application to cell typing in clinical transplantation.
