【Abstract】 The localization of Factor Ⅷ-relatcd antigen (Ⅷ R: Ag) in human splenic tissue using euzyme-linked immuno-sorbent assay with peroxidase-protein A (PPAELISA) is reported. The posititive brownish granules are unevenly distributed in the cytoplasma of the vascular endothelial cells. Special emphasis on the exploration of the method for removing non-specific endogcncous peroxidases was made, and the optimal concentrations of rabbit antihuman Ⅷ R: Ag serum (Ist antibody) and PPA (second antibody) for this test were established. The results were compared with those obtained from the indirect immunofluorecent staining method and proved similar. It is believed that this method is sensitive and simple for the localization of Ⅷ R: Ag and the specimen can be kept for a long period.更多还原
