色谱分析在菌群研究中的应用

【摘要】 为有助于菌群研究中厌氧菌和兼性厌氧菌的分类和鉴定,本文以GLC法分析了486株菌的代谢产物.比较了VPI和上野等提取代谢产物的方法,结果两种提取方法效果相当.两套分析系统[CAPCO(TCD)分析系统和GC-7AG(FID)分析系统]在分析菌的代谢产物方面各有所长,最好能并用.GLC分析菌的代谢产物对厌氧菌的分类和鉴定很有价值,是菌群深入研究的重要手段之一.在厌氧菌的色谱图中,主要出现B、IB、V、IV、C、IC等色谱峰.而需氧(兼性)菌仅出现A、P、L或有S,这提示GLC分析方法可能直接用于临床,便于临床厌氧菌感染的早期诊断.

【Abstract】 The metabolic products of 486 strains of various bacteria were analysed by GLC for identification and classification of certain anaerobes and facultative anaerobes in a survey of normal bacterial flora. We compared VPI’s method with that of Kazue Ueno’s, and it was revealed that the results obtained were similar, as each of the analytic systems mentioned above [ CAPCO anaerobic analytic system (TCD) and GC-7AG analytic system (FID)] each possess its own advantages. It might be better to use both of them simultaneously.It was found that analysing bacterial metabolic products by GLC yielded valuable information in identification and classification of anaerobes,and it was found to be one of the useful means by which we could carry out an intensive research of flora. In the chromatograms of non-sporing anaerobes, the main peaks of fatty acids appeared in the ranges of butyric, iso-butyric, Valeric, isovaleric, Capric & Isocapric acids, whereas the chromatograms of aerobes or facultative aerobes showed that the chief peaks appeared in the ranges of acetic, propionic, lactic,and sometimes succinic acid. The presented data suggested that analysing bacterial metabolic products by G1C could be directly used with specimens in rapid clinical diagnosis of anaerobic infections as used in early tentative diaginosis.
