登革4型病毒的单克隆抗体

【摘要】 用SP2/0小鼠骨髓瘤细胞与用登革4型病毒免疫的鼠脾细胞,在PEG—1000的作用下进行融合,获得了8株产生抗登革4型病毒单克隆抗体的杂交瘤细胞系。其中5株杂交瘤产生的抗体,对登革4型病毒具有荧光型特异性,1株对登革1型病毒有交叉反应;另2株对登革2型病毒有交叉反应。对上述8株杂交瘤分泌的抗体又进行了补体结合、空斑抑制中和及血凝抑制试验的鉴定,结果有2株是补体结合型特异,另4株有低度的中和活性。然后用3个荧光型特异的单克隆抗体对5个地方毒株进行试用的结果,进一步验证了它们的特异性。上述杂交瘤稳定传代7个月后,冻存于液氮中。

【Abstract】 Eight hybridoma cell lines were obtained by fusion of mouse myeloma cells and splenocytes from mouse immunized with dengue-4 virus. Among them,5 hybridoma-producing antibodies were type-specific for dengue-4 by indirect immunofluorescence assay,the other two hybridoma antibodies cross-reacted with dengue-2 virus, and another preparation cross-reacted with dengue-1 virus. Serological characterization of 8 hybridoma-producing antibodies were identified by complement fixation, plaque reduction neutralization, and hemagglutination-inhibition tests. Two monoclonal antibodies were type-specific in CFT, while the other 4 monoclonal antibody preparations had a low neutralization titer. Then, 3 type-specific (IFA)monoclonal antibodies were evaluated for their ability to identify 5 isolates by indirect immunofluorescence assay, and high specificity was obtained. These 8 hybridoma cell lines had already secreted antibodies for seven months in vitro and stored in liquid nitrogen for use in further studies.
