钙离子对肺泡巨噬细胞吞噬功能的影响
EFFECT OF Ca2+ ON PHAGOCYTOSIS OF PULMONARY ALVEOLAR MACROPHAGES
【摘要】 本文观察了在体外温育条件下Ca2+对大鼠和豚鼠肺泡巨噬细胞吞噬能力的影响。结果表明大鼠和豚鼠的正常吞噬能力无明显差异,Ca2+可增加大鼠和豚鼠肺泡巨噬细胞的吞噬能力。三氟啦嗪和酚妥胺阻断Ca2+受体蛋白——钙调素之后,Ca2+对肺泡巨噬细胞吞噬能力的增强作用不再出现,说明Ca2+的这种作用可能是通过细胞内钙调素来实现的。
【Abstract】 To evaluate the hypothesis that Ca2 + may enhance the phagocytic ability of pulmonary alveolar macrophage (PAM), PAMs were obtained from lungs of normal rats and guinea pigs by saline bronchoalveolar lavage, and phagocytic rate and phagocytic index of PAM to Staphylococci albus were determined. There was a marked increase in phagocytic index when Ca2 + was added. After Ca2+ was eliminated by EDTA, the phagocytic index of PAM decreased. To clarify the mechanism of calcium-enhanced phagocytosis, an antipsychotic agent trifluoperazine (TFP) or phentolamine was used as an inhibitor to block the calmodulin activity, Ca2+ alone showed enhancement effect, but Ca2++ regitine did not result in the increase of phagocytic ability.These findings suggest: 1) Ca2+ exhibits enhancement effect of phagocytosis for both rat and guinea pig PAM; 2) intracellular calmodulin plays an important role in calcium-enhanced phagocytosis.
