抗沙眼衣原体的单克隆抗体杂交瘤细胞株的建立

【摘要】 融合沙眼衣原体TE-55株免疫的小鼠脾细胞和小鼠骨髓瘤NS-1细胞,共获得5个分泌特异抗体的杂交瘤细胞株。其中杂交瘤AH6和1-H3经有限稀释法再克隆化培养6次;用ILISA法和RIA法测定抗体。核型分析和FACS测定表现出它们比NS-1细胞具有更多的染色体数和DNA含量。AH6和1-H3均已传代7个月以上,稳定分泌单克隆抗体,它们在小鼠体内很容易产生高效价腹水。另外讨论了某些增加融合成功率的重要条件。

【Abstract】 The five hybridoma cell lines secreting monoclonal antibodies against chlamydia were established by fusion of NS-1 mouse myeloma cells and limouse spleen cells immunised with C Trachomatis TE-55. Two of them, AH6 and 1-H3 have been subcloned for six times with limiting dilution procedures. Hybrid clones that produced specific antibodies were screened by ILISA and RIA. Karyotype and FACS analysis of AH6 and 1-H3 showed more chromosomes and DAN content as compared to that of NS-1 cells. AH6 and 1-H3 cell lines were stable to secrete monoelonal antibodies in tissue culture more than seven months. It is easy to make mice produce ascites with high valence by hybridoma cells, On addition, some important factors about increasing positive rate of fusion is discussed.
