自身骨髓移植研究之一——骨髓的保存

【摘要】 在恶性肿瘤的治疗中,国外目前应用强烈化疗或放疗,以期达到根治之目的.但其严重的骨髓抑制大大地限制了它的应用.自身骨髓移植就是在化疗前从人体内抽出,并在体外保存一段时间,然后待药物的毒性作用消失再将其回输入体内.这就避免了上述的副作用.因此能否有效地保存细胞,维持其活力和功能是自身骨髓移植的关键.本文介绍了骨髓的保存方法,尤其是超低温保存.取得的结果是令人满意的.人骨髓于4℃可有效地保存3天,有核细胞、锥蓝拒染率和粒系定向干细胞(CFU—C) 的回收率分别为84%、91%和97%.骨髓贮存于液氮中,其细胞的活力和功能得以较长保存,有核细胞、锥蓝拒染率、脱氧核糖核酸(DNA)和CFU—C的回收率分别为76%、89%、77%和79%.骨髓造血干细胞在体外成功的保存,为临床开展自身骨髓移植提供了先决条件.
【Abstract】 The intensive chemotherapy or radiotherapy has now been applied in the treatment of cancer abroad so as to get a radical cure, but its application is restricted because of the serious suppression of bone marrow. Bone marrow is drawn out before chemotherapy, preserved in vitro for some time, and after the effects of the medicine disappear, retransfused into the body. This is what autologous bone marrow transplantation ( ABMT ) means. Thus the above-mentioned harmful effects can be avoided. Therefore, whether or not we can effectively preserve cells and sustain its viability and function is indeed -the key to the successful use of ABMT. The article introduces the cryopreservation of bone marrow. The result attained is very satisfactory. The bone marrow of human beings is effectively preserved for three davs at 4℃. Recoverable rate of the nuclear, exelusion of trypon blue and colony-forming-unit culture ( CFU-C ) are 84%, 91% ar.d 97% respectively. When the bone marrow is preserved in liquid nitrogen, the viability and function of the cells can be sustained much longer. The recoverable rate of the nuclear cells, exclusion of trypon blue, DNA and CFU-C arc 76%, 89%, 77% and 79% respectively. Successful preservation of hematoiogic stem-cell in vitro provides predisposing factor fcr applying ABMT clinically.
