应用尼龙毛分离T、B细胞检测酯酶在淋巴细胞的分布(简报)

【摘要】 <正> 本实验用尼龙毛将经聚蔗糖—泛影葡胺分离的PBL分为粘附与非粘附部分,尔后分别用EtAET花结—酯酶、C3花结—酯酶两种双标记法分别测定,进一步分析酯酶在淋巴细胞中的分布.取正常肝素抗凝血4~5ml,用淋巴细胞分层液提出淋巴细胞,再以尼龙毛过滤,过滤柱应用结核菌素注射器,装填洁净的尼龙毛70mg,柱高6cm,以10%小牛血清—Hank’s液作营养洗液,收获的非粘附细胞主要为T细胞,粘附细胞主要为B细胞.前者用EtAET—酯酶双标法测定,后者用C3花结—酯酶双标法测定.

【Abstract】 This article presents a trilabelled assay of mixed Et ( AET )-C 3 rosette and acid-α-naphthyl acetate esterase ( ANAE ) of the examination of PBL of 26 normal individuals, E rosette with AET treated with SRBC was used for the identification of T cells and C3 rosette for B cells. With this mixed rosette assay PBL could fall into 4 types ( T,B,M and D cells ) in a single smear. The percentage and the distribution cf ANAE+ and ANAE-cells in 4 types were 71.5 ( ANAE + 51.6, ANAE-19.9 ) for T cells, 10.5 (+5.5, -5.0) for B cells, 16.5( + 6.9,-9.6)for N cells and 2.5(+1.9, -0.6)for D cells Most of ANAE+ cells were T cells, but not all there for ANAE+may represent a certain stage of development of lymphocytes.
