辐射交联聚乙烯醇水凝胶人工玻璃体的研制
RADIATION CROSSLINKED PVA HYDROGEL FOR VITREOUS IMPLANTATION
【摘要】 本文用电离辐射方法使聚乙烯醇在水溶液中交联生成水凝胶,凝胶剂量是温度和聚合物浓度的函数.在20℃ 0.6Mrad辐射交联7%PVA制得的水凝胶,经抽提可溶性PVA后,得无色透明、透光率97~99%、含水率>99%、比重为1.006、pH值为7的水凝胶。这些性能均与人眼玻璃体相近,是一种较理想的新型人工玻璃体。该人工玻璃体已用家兔做了70~80例的置换试验,手术初期有轻微炎症,两周后逐渐消失。大部分置换眼玻璃体透明、无界面、视网膜清晰可见、眼压正常。经置换的玻璃体最长已观察了600天以上。初步药理检验表明无毒性。
【Abstract】 <ABSTRACT>Crosslinked hydrogel prepared by irradiating on aqueous solution of polyvinylalcohol ( PVA ) was studied. It was found that the irradiation dose of gelation is a function of irradiating temperature and polymer concentration. Hydrogel, prepared by irradiating 1% aqueous solution of PVA for 0.6 Mrad at 20癈 in atmosphere nitrogen and extraction of soluble PVA, was a transparent, fluid gel, having a transparency of 97~ 99%, water content of 99%, density of 1.006 and pH of 7. These properties are similar to those of human vitreous and can be used for an ideal artificial vitreous.PVA hydrogel, implanted in rabbit eyes ( 70~80 rabbits ) appeared to be slightly imflammatory during the initial stage after implant operation and gradually returned to normal after 2 weeks. Majority of the implanted vitreous were transparent and without visual interface. Fine retinal structure could be seen through the hydrogel. Intraocular pressure of all eyes was normal for over 600 days. Initial pharmacological test showed that the hydrogel was nontoxic.
