微生物产生的酶抑制剂研究——Ⅰ.蛋白酶抑制剂的筛选方法探讨
STUDIES ON ENZYME INHIBITOR PRODUCED BY MICROORGANISM—— I. METHODS OF SCREENING FOR PROTEASES INHIBITORS
【摘要】 建立了一个简单的筛选胃蛋白酶、糜蛋白酶、胰蛋白酶及α-淀粉酶等抑制剂的平板透明图法。这个方法的基本要点是:1.平板是由琼脂和所筛选的酶的底物混合组成;2.测定时发酵液和酶是在酶解反应最适的条件下直接结合,在37℃保温反应30分钟,3.琼脂平板是用打孔器打洞,直径为9mm;4.反应混合物滴入洞内保温反应;5.经一定时间后观察、测定透明圈的有无或大小。本法既不同于日本武田研究所建立的平板法也不同于我国何继春等的平板法。它具有透明圈清晰、方法简易、酶用量少等优点。
【Abstract】 A simple method has been devised for the screening of inhibitors of pepsin, chy-motrypsin, trypsin and alpha-amylase inhib-itor. The features of the method are: l,the agar plate is mixed with substrate of the specified enzyme; 2, the culture filtrate and the enzyme are mixed under the optimum conditions of enzyme’s reaction. The reaction mixture is incubated for 30 minutes at 37℃; 3, the agar plate is plugged with cork borer to form a hole of 9mm in diame-ter; 4, the reaction mixture is dropped in to the hole; 5, after incubation at a given time, the result is observed. If there is no zone formed, the culture filtrate is supposed to contain the enzyme inhibitor. This method is different from plate methods proposed by Kakinuma A et al and of He Ji-chun(何继春). The vitreous zone obtained by our method is clear. This method is simple and quantity of enzyme used is small.
