抗真菌抗生素414的鉴别

【摘要】 抗真菌抗生素414是一个芳香七烯大环内酯类的抗生素。由于这类抗生素都是结构复杂的、多组分的复合物,它们难溶于水和一般有机溶剂、稳定性差,这些都极不利于它们的分离和鉴别。本文利用螺旋管行星式逆流层析法和高效的反相液体色谱法,直接分离比较抗真菌抗生素414和相关的七烯抗生素。证明在主要的组成组分上,抗真菌抗生素414和杀假丝菌素是同质的。研究比较了这二个抗生素的一系列光谱学特征,也支持了抗真菌抗生素414和杀假丝菌素同质的结论。

【Abstract】 Antifungal antibiotic 414 is produced by Streptomyces globoroseoviolaceus n.sp.isolated from soil samples collected in Nanchang, Jiangxi province, China.It belongs to Cand-icidin subgroup of heptanenes, because of it contains mycosamine and has a sidechain of p-aminoacetophenone in its molecule.Examination by means of the coil planet centrifuge and by reverse phase HSLC, it was clearly indicated that identity of the major components of antifungal antibiotic 414 complex and Candicidin complex, whereas the major components of Trichomycin was clearly different from those of antifungal antibiotic 414 or Candicidin complex. The conclusion was further supported by theadditional UV., IR and PMR studies.
