【摘要】 本文对ELISA间接法检测HSV抗体作了下列研究:以两种较简易的方法,即病毒感染细胞连同维持培养基一起(方法一)或经洗涤后在PBS中(方法二)反复冻融,分别从原代兔肾、原代鸡胚、传代Vero和FL细胞制备HSV抗原,结果以方法二制取的Vero细胞病毒抗原滴度最高;原代兔肾者次之。选取16份ELISA呈不同滴度的血清作中和试验,两法结果呈正相关关系(r=0.79,P<0.005)。初步得出△A492≥0.2可作为血清抗体阳性。以ELISA检测3组年龄人群血清抗体,婴儿组抗体阳性率6.6％,儿童组80％,成人组100％。最后,比较了酶联抗人IgG及酶联SPA ELISA检测的29份血清抗体含量,两种结合物测得的结果呈正相关关系(r=0.986,P<0.0005),并提出酶联SPA在鉴别两型HSV血清中具有重要意义。更多还原

【Abstract】 This paper reports the study on the ELISA indirect method for detection of antibody to HSV as follows: The HSV antigens were prepared from the cells of the primary rabbit kidney, primary chicken embryo. FL and Vero by two simple methods i. e. freezing and thawing the virus-infected cells repeatedly either with maintaining the medium ( Method Ⅰ ) or after being washed with PBS ( Method Ⅲ). It was found that the titer of antigens prepared from Vero cells by method Ⅱ was the highest and that from primarv rabbit kidney cells was lower. Antibody titers in ELISA from 16 selected sera were compred with the antibody titers by the neutralization test and a significant relationship (r=0.79, P<0.005 ) was found between them. The ELISA AA492>0.2 was considered positive. The levels of antibody in three age groups were detected with ELISA. The findings showed that the positive rate of the infant group was 6.6%, of the children group 80% and of the adult group 100%. Finally, the comparison of enzyme-labelled anti-IgG with enzyme-labelled SPA in ELISA for 29 sera was performed, The results have indicated that the value obtained in SPA-ELISA is close to that in anti-IgG ELISA (r=0.986, p<0.0005). It suggests that the use of enzyme labelled SPA is significant in differentiating serum antibodies into HSV type Ⅰ and Ⅱ.更多还原
