枇杷茎尖培养与增殖的研究
A STUDY ON TISSUE CULTURE AND INDUCING PROLIFERATION FROM SHOOT APEXES OF LOQUAT TREE ( Eriobotrya japonica Lindl. )
【摘要】 本文报道了不同来源的枇杷茎尖离体培养成功的方法与条件。取自实生幼树的茎尖在MS+6-BA0.5+NAA0.1+GA3 0.1~0.2(毫克/升)培养基上顺利展叶,而后转接到MS+6-BA0.25+IAA0.05(毫克/升)中抽枝,在无激素的1/2MS培养基中长根。而取自嫁接结果树的茎尖则需要更高的外源激素。部分品种可在上述培养基中展叶,但必须在MS+6-BA1.0+NAA0.25(毫克/升)的培养基中才能抽枝,它需要用100毫克/升的IBA处理12小时才能长根。谷胱甘肽在枇杷茎尖离体培养中作用不大,而初期的全黑暗培养可提高成功率。不同生长状态的芽培养成功率各不相同。 本文还探讨了影响枇杷茎尖苗增殖速度的条件,认为6-BA可以抑制顶端优势,促进腋芽萌发,根据新枝增殖率和正常率与6-BA浓度之间的数量关系,认为最适繁殖的6BA浓度为1.5毫克/升。
【Abstract】 This study reports a successful method of tissue culture and inducing proliferation from the shoot apexes of different varieties of loquat tree( Eriobotrya japonica Lindl. When the shoot apexes from loquat seedlingswere cultured in a MS medium with the addition of 6BA 0.5,NAAo.l,and GA3 0.1~-0.2(mg/l), they grew well to expand their leaves Afterthey were transferred into a medium of MS + 6BA 0.25 + IAA 0.5( mg/1 ) they grew into shoots and became young seedlings. But their ro-ots were formed in the other medium, i. e. 1/2 MS without the addition of any hormone. Nevertheless, the shoot apexes from the grafted fruit-setting trees need more external source hormone. Some loquat varieties can expand their leaves in the same medium given above, but they only developed their shoots and became young seedlings in the medium of MS+ 6BA 1.0 + NAA 0.25 (mg/1), and produced their roots by the treat-ment with IBA 100mg/l for 12 hours. The experiment showed that the fu-nction of glutathione was not significant in the culture of loquat shoot apex, and the initial stage of culture in full darkness can increase the rate of success. However, the rate of success varies from different grow-ing state of the buds used in the culture. Furthermore, the condition that effects the proliferation rate of loquat shoot apex was also discuss-ed in this paper. It was shown that 6BA inhibited the apical dominance and promoted the axillary bud to grow. According to the quantitative relationship between the concentration of 6BA and the rates of prolifer-ation and normality, the optimum concentration of 6BA for propagation was 1.5 mg/1.
