菜白蝶的一种新病毒——Ⅱ.病毒的生物物理、血清学性质
A NEW INSECT VIRUS OF PIERIS RAPAE BIOPHYSICAL AND SEROLOGICAL PROPERTIES OF THE VIRUS
【摘要】 用氯仿-丁醇处理和差速超速离心,再经两次蔗糖密度梯度离心,纯化了这株菜白蝶病毒。病毒沉降系数约132S。在氯化铯中的浮力密度为1.39g/cm~3。在相同条件下测定,健康菜白蝶幼虫蛋白沉降系数分别为16S、38S、76S,而被病毒感染的死幼虫则为17S、62S、127S。纯化的和部分纯的病毒与抗部分纯病毒血清,在0.9%琼脂糖中双扩散反应时,纯化病毒出现一条沉淀线,并与部分纯病毒两条沉淀线之一的两端相融合。当粗提取的健康幼虫蛋白和纯化的病毒与抗部分纯病毒血清反应时,纯化病毒产生一条独立的沉淀线,而不与健康幼虫蛋白沉淀线融合,呈现典型的不一致反应。
【Abstract】 A new insect virus of Pieris rapae was purified with chloroformbutanol treatment followed by differential centrifugation and sucrose gradient centrifugation. The sedimentation coefficient of virion was approximately 132S, and it banded at a density of 1.39 g/cm~3 in CsCl.The sedimentation coefficients of the protein components from the healthy larvae were 16S, 38S, 76S, while those from dead infected larvae were 17S, 62S, 127S respectively. The last component (127S) contained the virus.Both purified virus and partially purified virus preparations from dead infected larvae gave a corresponding precipitation line in gel when tested with the antiserum against partially purified virus. When crude extracts of healthy larvae and purified virus were tested with antiserum against partially purified virus, the pure virus produced a precipitation line which did not join to that formed by healthy extract.
