γ辐射对红细胞膜流动性的影响

【摘要】 我们实验以红细胞膜作为材料,使用ANS和DPH荧光探针测量红细胞膜的荧光强度和偏振度。发现γ辐照红细胞膜剂量从4.6×10~3拉德增至148×10~3拉德,而ANS和DPH的荧光强度随剂量的增加而降低,荧光偏振度随剂量的增加而增加,这些结果表明γ辐照引起红细胞膜流动性降低。但荧光强度的减少并不伴随荧光光谱的变化。ANS与γ辐照的膜结合量也没有改变,因此荧光强度的减少可能是由于在辐照的膜中ANS荧光量子产率的降低。在我们的实验中γ辐射对ANS探针所引起的荧光变化较DPH灵敏,看来γ辐射首先影响膜蛋白。

【Abstract】 Erythrocyte membranes were usedas a material in our experiment.Fluorescent dyes,such as 1-anilino-8-naphthalene sulfonate (ANS) and1,6-diphenyl-1,3,5-hexatriene (DPH)were used as probes to examine thefluorescence intensity and polarizationof erythrocyte membranes.It wasfound that the fluorescence intensityof ANS and DPH decrease in γ-ir-radiated membranes with increasingγ-ray doses from 4.6×10~3 rads to148×10~3 rads.But the fluorescencepolarization of ANS and DPH incor-porated into the membranes indicatedthat its degree of polarization increa-ses.These results show that γ-ir-radiation can induce a reduction offluidity of erythrocyte membranes. The fluorescence decrease was notaccompanied by a change of shapein the fluorescence spectrum.Theamount of ANS combined wih γ-ir-radiated membranes was not changed.The fluorescence decrease might be dueto decrease in quantum yield of ANSfluorescence in the irradiated membranes.In our experiment the changein fluorescence of ANS probe cause by.γ-irradiation was more significant thanthat of DPH.It seemed that γ-ir-radiation primarily affects the membrane protein.
