血清甲状腺素T4固相放射免疫测定
SOLID PHASE RADIOIMMUNOASSAY FOR THYROXIN(T4)
【摘要】 T4固相放射免疫方法是近几年来国外发展的一种新的放射免疫技术。我们研制了用聚甲基丙烯酸甲酯12×60mm塑料管和聚苯乙烯小球(直径6.35mm),二者均进行特殊加工而成,并通过化学联结方法,将T4抗体结合在上述固相载体上作为抗体和抗原竞争免疫反应的吸附剂,从而简化分离步骤,反应时间1小时,取血样10~50μl。与PEG法、双抗体法、CPBA法进行了比较,T4固相法更为稳定、快速、方便。T4固相小球法和固相管法,正常值分别为8.53(μg/dl)、9.33(μg/d1)。并进行了2,000例病人考察。此固相技术还可应用于多肽激素、甾体激素以及一些蛋白激素的测定。
【Abstract】 The measurement of thyroxin (T4) hormone by radioimmunoassay has been facilitated by the introduction of -solid-phase antibody preparations which permit the separation of bound and free tracer to be performed by a rapid and convenient method. We have used chemical promoter methods for coupling antibody to plastic beads and tubes. Only 10-50 μl of the test serum sample was needed and the time for reaction was one hour. As compared with other methods for example, PEG, double-antibody and CPBA, solid-phase method was more stable.By using this solid-phase method, the normal value of mean T4 concentratiions (μg/dL) measured were as follows: coated-beads 8.33, coated-tubes 9.33. The methods of using coated beads and tubes are simple, rapid and reliable and they can be in general, applied to assay of other ligands, such as polypeptide hormones, steroids and other protein molecules.
