不同温度下水牛和奶牛精子微细结构与细胞外酶活性的变化
CHANGES IN STRUCTURE AND EXTRACELLULAR ENZYME ACTIVITY OF WATER BUFFALO AND DAIRY BULL SPERMATOZOA UNDER VARIOUS LOW-TEMPERATURES
【摘要】 在不同低温下,就水牛和黑白花奶牛中精子的活力、顶体结构和细胞外 GOT、GPT 及 LDH 活性的变化进行了比较研究。根据海子水牛和黑白花奶牛各4头精液的测定,经冷休克及冷冻的水牛与奶牛精子活力的变化(包括解冻后活力)基本无差别,但冻后水牛精子的正常顶体率比奶牛少28.9%。50%的顶体损害均发生在冻前阶段。冷却至5℃平衡对稀释后一小时的水牛精子正常顶体率虽影响不大,但有约30%的受损害顶体变得严重肿胀,而对奶牛精子受损害的顶体并无作用。经过稀释、冷却和冷冻,水牛精清中的 GOT 活性,奶牛精清中的 GOT、GPT、LDH 活性逐渐增加,而且有显著差异,但水牛精子释放 GOT、LDH 的程度高于奶牛精子。冻后精子活力、正常顶体率、精清中酶活性之间的相关性较低,但水牛精清中 LDH 活性在冷冻期间的变化与冻后精子活力相关极显著。海子水牛冻精超薄切片的电镜观察表明,顶体外膜的“空泡化”过程明显,细胞膜本身发生膨胀与裂解,而未能证实形成“杂合泡”的现象。此外,尚见到类似“结节状”精子的顶体肿胀形态,可能意味着水牛精子顶体对低温处理的敏感性。
【Abstract】 A Comparative study of effects of various low-temperature conditions onthe changes in motility.Structure and activities of some extraeellular enzymesof water buffalo and cattle spermatozoa were carried out.From determining thesemen samples of each individual of 4 Chinese water buffaloes(Hae-Zhi breed),2 Murra buffaloes and 4 Black-white dairy cattle.The results observed wereas follows.There were no significant differences in the changes in motilityincluding the motility of post-thawing semen of buffalo and cattle.However,the normal acrosome rate of post-thawing water buffalo semen was less 28.9%than that of cattle.50% abnormal aerosomes occured in the stage before freez-ing.Cooling to 5℃ and equilibration for 5 hours didn’t decrease the normalacrosome rate of buffalo and cattle semen that had diluted and kept 1 hourunder room temperature.By the same procedure,about 30% acrosomes of buffalospermatozoon which had impared before cooling became swellen severely,butdidn’t affect the damaged acrosomes of cattle spermatozoon.The activity of GOT in the buffalo seminal plasma and the GOT、GPT andLDH activities in the cattle seminal plasma increased gradually during dilutioncooling to 5℃ and after freezing.The differences in activity of the enzymeswere significant.Nevertheless,GOT and LDH content leaked from buffalospermatozoon were higher than that from cattle.Among the post-thawingmotility,the normal acrosome rate and the activity of extracellular enzymes,no significant relationship was observed,but there was a positive relationshipbetween the increase in the LDH activity of buffalo seminal plasma afterfreezing and the post-thawing motility.Electron microscopy study on the superstructure of buffalo spermatozoonin the frozen semen demonstrated the apparent vesiculation at outer acrosomemembrane,the swelling and breakage of plasma mambrane.The so-called“hybridvesiele”havn’t been observed in this study.A swellen acrosome of buffalo sper-matozoon similar to the“Knobbed”bull spermatozoa has been noted and mayimply the susceptibility of the acrosome ridge of Chinese water buffalo sper-matozoon to low-temperture.
