【Abstract】 Two monoclonal antibodies raised against HSB-2 cells were produced and characterized. Both of them were IgG1 subclass antibodies and termed H65 and H154separately. They were reactive with 3 T cell lines, peripheral E~+ lymphocytes and thymocytes,but unreactive with RBC, B and other cell lines so far tested by binding assay. With indirect immunofluorescence, it was found that H65 and H154 reacted with more than 80% of peripheral E~+ lymphocytes as well as a substaintial percentage of thymocytes. A primary obaervation indicated that they also reacted with B-CLL cells.As anti-Leu-1 antibody,H65 and H154 recognized a T cell surface antigen with molecular weight 67K. Furthermore, the results of adsorption and blocking experiments showed that these three antibodies recognized the same molecule but different antigen determinants. The molecule defined by these three antibodies can be induced to increase in amount by TPA. This confirms the observation by others that the antigen is a mature T cell differentiation antigen. So that it is obvious that H65 and H154 are anti-Leu-1 like antibodies.更多还原
