雌激素对小鼠骨髓粒系定向祖细胞集落产率的影响

【摘要】 本文介绍了用“体外琼脂培养技术”和“体内扩散盒琼脂培养技术(ADC 法)”观察雌激素对小鼠骨髓粒系定向祖细胞集落产率(VCFU-C)的影响。体外培养的结果表明,雌激素对小鼠股骨骨髓中 CFU-C 的增殖具有抑制效应。

【Abstract】 The effects of estrone and estriol on the proliferation of marrow granulocytecommitted progenitor cells of normal mice are discussed in the paper.Progenitor cells in mouse bone marrow were assayed through their progeny inplate culture for 7 days.Both estrone and estriol have shown inhibitory effects onCFU-C(Colony Forming Unit Culture).When estrone with concentrations rangingfrom 10-12M,10-8M to 10-4M,and estriol with concentrations ranging from10-13M,10-9M to 10-5M,were added to the plates,there showed striking decreaseof CFU-C in the culture plate.The inhibitory rate of each concentration was54.4%,50.9%,61.6% and 33.1%,36.3%,43.5% respectively during 7 days ofculture.The inhibitory effects of estrone and estriol were similar.During 5 days ofculture by mean of agar diffusion chamber (ADC),CFU-C Showed a 26.2% increasein the castrated mice compared with the control.The present work demonstrated that estrone and estriol exert inhibitory effectson CFU-C in Vitro and it is possible that estrogens have inhibitory effect onCFU-C in Vivo ADC.The latter needs further studying.
