多肽激素的酶促125碘化及其保存稳定性研究

【摘要】 <正> 作者们自1978年起迄今三年间先后完成了HTSH、HGH、HCG、HLH、和OFSH等五种多肽激素的乳过氧化物酶(Lactoperoxidase,简称LPO)酶促125碘化法,结合国内目前供应的125碘化钠(含还原剂)的具体条件,选择酶促125碘化的最佳条件;酶及其组分内杂蛋白的自身125碘化问题;并对酶促法与氯胺T法制备的多肽激素的保存稳定性进行了对比研究,现总结如后。

【Abstract】 In this paper, enzymatic radioiodination of five polypeptide hormones (HTSH, HGH, HCG, HLH, OFSH) accomplished since 1978 is described studies were carried out in three respects: (1) The influance of various experimental conditions on enzymatic radioiodination, such as, amounts of LPO and H2O, pH value, time and temperature of reaction etc; (2) Stability of polypeptide hormones in storage and comparison between LPO method and CT method. It was shown that immunoactivity of former method is better and move persistant than that of the latter; (3) Beyond polypeptide hormones, radioiodination of LPO were studied. With practical amount of LPO, the iodination of LPO was trifle.
