应用冷冻蚀刻技术研究几种病毒的超微结构

【摘要】 采用冷冻蚀刻技术研究了GPV、IBRV和NDV等几种病毒的超微结构,初步取得如下结果: 一、在冷冻蚀刻样品中,GPV呈球形(在细胞质内)或近似球形(释放到细胞外),直径约300nm,这与在超薄切片或负染色样品中所见的病毒形态和大小有很大的不同。二、IBRV囊膜的PF和EF面上,均可见到直径5—9nm的膜内微粒。GPV囊膜膜内微粒的数量及其在PF面和EF面上的分布与IBRV不同。细胞质内与细胞外成熟的GPV囊膜的膜内微粒也有明显的差异。三、在GPV毒浆基质中,可观察到由直径5—10nm颗粒的聚集或规则排列形成的弓形、环形、及直径150—400nm大小不等的球形结构。实验结果显示了冷冻蚀刻技术在病毒形态结构和形态发生,尤其在病毒囊膜结构研究中的优越性。对这一技术在病毒学中的应用进行了讨论。

【Abstract】 The fine structure of GPV, IBRV and NDV were studied by means of freeze-etch-ing electron microscopy. Some initial results were obtained as follows: 1. Freeze-etching revealed that the mature GPV appear to be spherical in cytop-lasm or oval after released from host cell and have a diameter of approximately to 300nm. These are quite different from those seen by thin-sectioning and negative stainingtechnique. 2. Intramembrane particles measuring 5-9 nm in diameter can be seen on both Pand E faces of IBRV envelepes. Their member and distribution on P and E faces aredifferent from those of GPV. Moreover the intramembrane particles of GPV envelopematured in cytoplasm are quite distinguishable from that released from host celis. 8. A lot of particles, 5-10 nm in siz% aggregate in the viroplast matrix of GPV.Sometimes the aggregates appear in arched, circular or spherical form with a diameterof 250-400 nm. Freeze-etching technique is favorable to study the viral morphology and morphogene-sis, especially on viral en~,elopes. The applieatiqn of this technique in virology is dis-cussed.
