苯中毒时骨髓细胞功能的实验研究
Study on the Functions of Bone Marrow Cells in Benzene Poisoning
【摘要】 本实验采用~3H-TdR参入-高速放射自显影和脾结节生成技术等现代实验血液学方法,对苯中毒时骨髓细胞的功能变化进行了动态观察,并比较了不同苯剂量对其骨髓毒性的影响。 结果表明,轻度苯中毒时,其骨髓毒性可能主要由于骨髓分化细胞DNA的合成受到干扰而引起,骨髓造血微环境可能也受到波及。 本文还讨论了苯中毒的临床发病机理。认为苯中毒可能会损及包括造血干细胞和造血微环境在内的骨髓所有成分;苯的作用强度则可能是此种毒性作用的关键,使得苯中毒具有延续性、多样性和可逆性的临床特点。
【Abstract】 Modern experimental hematology methods such as ~3H-TdR incorporation with high-speed autoradiography and spleen colony-forming technique were used to observe the changes of bone marrow cells’ functions in benzene poisoning mice, and a comparison of effe.cts was made between the two different dosages of benzene on the myelotoxicity.The results revealed that in mild benzene poisoning the myelotoxicity seemed primarily due to the depression of the repopulation ability of differentiated bone marrow cells by the interference in DNA synthesis, and probably the hematopoietic inductive microenvironment (HIM) was also involved. The pathogenesis of benzene intoxication was discussed. It was considered that benzene poisoning may influence all bone marrow elements including hematopoietie stem cells and HIM, and that the keypoint of this mechanism was the action intensity (dosage and time) of benzene which caused the benzene myelotoxicity characteristics of continuity, variety and reversibility.
