胚鼠大脑皮层神经细胞的体外原代分离单层培养方法
The Primary Dissociated Monolayer Culture of Developing Mouse Cerebral Cortex Neuronal Cells.
【摘要】 我们按照Swaiman氏方法建立胚鼠大脑皮层神经细胞原代分离单层培养。细胞可在体外存活20天以上并具有特异的形态特点及3H—GABA摄取功能。如将细胞接种到16毫米直径小皿中在体外存活后,可获得较大数量而条件近似的样品而适宜于神经生化和神经药理的研究。对于进一步在细胞和分子水平上研究发育中的脑的问题而言,这种类型的神经细胞培养具有一定实用价值。
【Abstract】 The primary dissociated monolayer culture of developing mouse cerebral cortex neuronal cells was established. Cells can be alive in vitro for more than 20 days and possess specific morphological and ~3H-GABA high affinity uptake character identified by autoradiography. Seeding cells to the cluster wells of 16 mm diameter provide relatively large-scale samples with very close condition quantified by Lowry protein test and ~3H-GABA uptake utilizing liquid scintillation counting.This model is available for the research about neurochemistry and neuropharmacology. Moreover, in order to study the developing brain at a cellular or molecular level, neuroronal cell culture of such kind is of some valued.
