应用吖啶橙免疫荧光法快速检定痢疾杆菌的报导

【摘要】 本文对145例拟诊为菌痢患者的粪标本,采用吖啶橙荧光抗体(AOFA)法进行检定,同时以常规培养法作对照,两种方法所得结果的阳性符合率为90%.本法特异性强:痢疾杆菌AOFA只对痢疾杆菌形成特异性的荧光菌团,且能从此荧光团中分离出典型痢疾杆菌,对不凝集弧菌等其他肠道12种细菌皆不形成典型荧光菌团.敏感度高:每ml含1000~10000以上细菌呈“+”~“卌”的荧光菌团.因此可认为AOFA法是一种特异、敏感、快速、简易、节省材料等的诊断方法,而且可从荧光菌团中分离培养到活菌作确定诊断.

【Abstract】 Stool specimens of 145 patients clinically diagnosed as bacillary dysentery are examined by using acridine orange-fluorescent antibody (AOFA ) method and routine culture in controls. It is found that the identity rate of positive reactions obtained by these two methods is 90%. AOFA of bacillary dysentery is specific to the bacteria in forming fluorescent clumps, which could not be rendered by tests carried out with nonagglutining vibrio and enteric bacteria of 12 species. Furthermore, the viable bacteria may be isolated by immune-fluorescent clumps and be incubated for identification. AOFA gave a higher sensitivity in sample containing bacteria number over 108~10~4/ml, it did showstrong positive reaction ( +-→++++) of immunofluorescent bacterial clumps.This test may be used as a rapid, specific, sensitive, comparatively simple and less expansive method in determining bacillary dysentery.
