酶联免疫吸附法(ELISA)检测豆类种传病毒的研究
STUDIES ON THE DETECTION OF SEED-BORNE VIRUS BY ENZYME-LINKED IMMUNOSORBENT ASSAY (ELISA)
【摘要】 本文介绍使用微量板酶联免疫吸附法(ELISA)检测TMV(杆状)、SMV(线状)、CMV(球状)、AMV(杆菌状多粒体)四种不同形态、不同分类组群的病毒。此法非常灵敏,能够检测病毒提纯液的最低浓度为7.6~10亳微克/毫升,也能检出稀释1563~10240倍病叶粗澄清液中的病毒,灵敏度比琼脂双扩散法高1000倍以上。 ELISA可以直接检出单粒种子中的病毒,而且可以分别检出单粒种子不同部位如大豆的种皮、种胚、胚乳及胚根中的病毒存在。该法适用于检测大量标样,对植物检疫来说,这是一个很好的方法。
【Abstract】 The microplate enzyme-linked immunosorbent assay (ELISA) of determining the viruses of four different shapes (rigid red,long flexible thread,isometric and bacillus-like) and of different groupes was described.With a high sensitivity,it can determine the clarified virus of minimum cencentration at 7.6-10ng/ml and also determine the virus of the diseased leaf sap being diluted to 1563-10240 times.The sensitivityof this method is 1000 times higher than that of the agardouble diffu-sion test. The virus in a single seed and in various parts of a single seedchsnas skin germ,endosperm,and radicle of soybean seed could be detectedby ELISA.It can be used on large-scale samples,and is a very goodmethod applied to plant quarantine.
