小鼠肺癌癌变研究(光镜、组织化学和超微结构观察)

【摘要】 黄曲霉菌和二乙基亚硝胺都能以昆明种小白鼠肺脏为靶器官,诱发出肺腺癌。生癌率分别是45.5%和71%。癌的发生部位在细小支气管及其末梢部位,来源细胞是支气管的无纤毛上皮细胞,即 Clara 细胞。靶细胞在损伤通过的修复性增生、单纯性灶性增生和不典型灶性增生,继而成癌外侵。黄曲霉菌诱癌所需时间约为二乙基亚硝胺的2倍。除二乙基亚硝胺实验组癌细胞中有针状结构外,各种瘤细胞浆形态、代谢和超微结构类似。

【Abstract】 Pulmonary adenocaricinomas in a Kunming strain mice were induced with di- ethylnitrosamine(incidence 71%)or Aspergillus flavus link(incidence 45.5%)iso- lated from foodstuffs in Hebei Cixian County,an area of high cancer incidence.The induction period for Aspergillus flavus link was twice as long as that for diethyl- nitrosamine.The carcinogenic process studied by light microscope,translation elec- tron microscope and histochemistry revealed that the adenocarcinoma arose from the Clara cells in the terminal bronchioles and as results of simple and atypical hyper- plasia.In the hyperplastic cells,undergo hyperplastic change succinate dehydrogenase activity in cytoplasm and nuclear deoxyribonucleic acid were found to be increa- sed.The glucose 6-phosphate dehydrogenase activity was higher in tumors than in normal lung tissue.In addition to needle-like structure in cytoplasm of cancer cells induced by diethylnitrosamine,there were abundant ribosomes,mitochondria,rough and smooth surface endoplasmic reticulum and Golgi complex.These carcinogenic changes were similar in those induced by either agent.
