水仙组织培养的研究

【摘要】 用福建漳州水仙(Narcissus tazetta var.chinensis)田间生长的和收获后贮藏的鳞茎为材料,使用加有一定激素的MS(1962)培养基。鳞茎材料分成两部分,一部分置于4~10℃低温下预处理4~9周;另一部分作对照。将鳞茎切成带有部分鳞茎盘的鳞片小块(长2~3毫米,宽1~2毫米),即是外植体。将外植体进行培养,结果表明,经低温预处理的,绝大部分外植体都可直接诱导成小植株。外植体的诱导率可达90~100%;未经预处理的鳞茎外植体的诱导率极低(2~6.67%)。我们认为,低温预处理是提高福建漳州水仙外植体诱导率的有效方法。

【Abstract】 Chinese sacred-lily from Zhangzhou, Fujian ( Narcissus tazetta var. chinensis ) was used as experimental material. The bulbs were obtained from the field when they were growing and from the storehouse when they were storing. MS ( 1962 ) medium supplemented with or without certain hormones. The bulbs were divided into two groups, one group pretreated in low temperature ( 4 ~10℃ ) for 4 ~ 9 weeks,and another group not pretreated as check. Then the bulbs were cut into segments which was 1 ~ 2 mm wide and 2 ~ 3 mm long each with a portion of the basal plate of scale, called explant. The result showed that the explants pretreated in low temperature could be mostly cultivated and induced to plantlets in MS(1962)medium. The rate of induction was 90~100%,while that of unpretreated check was very low(2.00~6.67%). Therefore pretreatment in low temperature is an efficient method to ob-tain high rate of producing bulblets or plantlets for Chinese sacred-lily.
