四季海棠的花药培养

【摘要】 作者用四季海棠(Begonia semperflorens Link et Otto)单核晚期花粉的花药进行培养,并获得了花粉植株。试验表明,诱导愈伤组织频率较高的培养基为B5+激动素1毫克/升+2,4-D2毫克/升+蔗糖5%;诱导愈伤组织分化苗较好的培养基为MS+玉米素10毫克/升+吲哚乙酸0.5毫克/升+蔗糖 3%(但玉米素浓度要逐步提高)。将分化形成的无根苗扦插在蛭石基质上,浇入一定量加有0.1毫克/升IAA的MS减半培养液,两周后即可生根。分化的植株经根尖染色体检查2n=x=18,证明为单倍体花粉植株。

【Abstract】 Studies on culture in vitro of Begonia semper florens Link & Otto ( 2n = 36 ) anthers containing microspores at late uriinucleate stage have been made, and pollen plants obtained. The experimental results show that B5 medium is superior to MS medium in the production of calli, and the highest frequency of callus induction is on B5 medium supple-mented with KT 1 mg/1, 2,4-D 2 mg/1 and 5% sucrose. The MS me-dium with various additions ( ZT, BA, KT, NAA and IAA ) was used for inducing differentiation of the calli, and the best result for inducing formation of shoots was found to be on the MS medium added with 10 mg/1 of ZT, 0.5 mg/1 of IAA and 3% sucrose ( concentration of ZT should be increased by degree ) .The formed shoots were isolated from calli and transplanted in the vermiculite medium watered with 1/2 MS nutrient solution containing 0.1 mg/1 IAA. The shoots transplanted in the vermiculite medium rooted after two weeks. Squash preparations of the root tips revealed that the chromosome number being 18 and demonstrated that the plantlets dif-ferentiated from the calli originated from the pollen grains.
