泡桐茎、叶的离体培养

【摘要】 作者报道了兰考泡桐(Paulownia elongata S.H.Hu)离体茎、叶的脱分化和植株再生。利用组织培养方法快速繁殖泡桐良种,是切实可行的捷径。 采用嫩梢、萌蘖芽伸长的嫩茎或分化苗叶片为外植体。在以MS为基本培养基,附加6BA(5~10ppm)和NAA(0.1~1ppm)的培养基上,得到了愈伤组织及再分化出的小植株。其中,以MS附加5ppm6BA和0.5ppm NAA的组合,分化率最高,获得的离体小植株也最多。 当小植株生长到1.5~2厘米时,切下置于1/2MS附加IBA(0.1~0.5ppm)的培养基内。约一星期开始生根。20~25天后,移栽到蛭石与草炭1:1的混合基质中。培养30天,可移至大田栽培。

【Abstract】 This paper describes the induction of calli and plantlets from stem and leaf culture in vitro of Paulownia elongata.The young shoots and young stems from sprouting buds or leaves of differentiated seedlings were used as explants. Calli and plantlets were obtained on MS basic medium supplemented with 6BA (5 to 10 ppm ) + NAA (0.1 to 1 ppm). The highest induction frequency of plantlets and the highest number of plantlets occurred on medium added with 5 ppm 6BA and 0.5 ppm NAA.The new growth of plantlet was cut off when it reached 1.5 to 2cmin height. Then transterred the plantlets onto medium with 1/2MS + IBA(0.1 to 0.5 ppm). About a week later they began to root; after 20 to25 days, transplant them into the mixed medium consisting of vermicu-lite and peat (1:1); 30 days later, transplant them to the field.This experiment had proved that by tissue culture, the time needed to produce plantlets was short, the coefficient of multiplication was big, the survival of plantlets was high. So, it is a practical method for rapid multiplication of stock trees in Paulownias.
