【摘要】 从胜利油田的油污土样中分离出一株革兰氏阳性、无芽孢、不分枝、不运动、不抗酸、能从原油产生胞外多糖的杆菌D9004。生长12—24小时,细胞0.5—0.6×1.5—3.5μm,到48小时,成为类球状和短杆。在营养琼脂平板上,菌落圆形,隆起,桔红色,表面光滑、润泽,边缘整齐。该菌不还原硝酸盐,石蕊牛奶中产碱,产生硫化氢,脲酶和过氧化氢酶阳性。从葡萄糖、果糖和蔗糖氧化产酸。不液化明胶。在SSC系统中,DNA解链温度(Tm值)为94.35±0.59℃,G+C含量为61.1±1.44克分子%。上述特征与黄橙色棒状杆菌(Corynebacterium flavo aurantiacum)基本一致。该菌以原油为碳源、摇瓶培养5—6天,发酵液粘度在35℃下为26cSt左右,总糖含量约3.5g/l。提纯的多糖经凝胶电泳和超离心分析证明为均一的。其沉降系数、特性粘度、偏比容、分子量和旋光度分别为6.28S,0.46dl/g,0.67ml/g,275,000和αD13+18。用纸层析法检出,该多糖与已报道的其它胞外多糖不同,由D-甘露糖、D-半乳糖(克分子比为2∶1)和丙酮酸所组成。其红外光谱与其它多糖的相似。

【Abstract】 A strain of bacterium D9004 producing extracellular polysaccharides from crude oil as C-source was isolated from oil-contaminated soil at Shengli oil-field. The organism is Gram-positive, nonmotile, non-sporulating, not acid fast, and unbranched. The cells growing for 12—24 h are 0.5—0.6 by 1.5—3.5 μm, for 48 h changed into coccoid and short rods. Colonies on nutrient agar plate are circular, raised, edge entire, orange-red, smooth, and moist-glistening. Nitrate is not reduced. Litmus milk is alkalified. Hydrogen sulfide produced. Gelatinenot liquefied. Urease and catalase: posi- tive. Glucose, fructose, and sucrose are oxidized into acid. The G+C content of DNA determined in SSC-system is 61.1±1.44 mol %, Tm value is 94.35±0.59℃. According to taxonomic studies this strain is identified as Corynebacterium flavo-aurantiacum.The organism produced about 3.5 g/l of polysaccharide after 5—6 days shake culture and the viscosity of the fermented solution was up to 26 cSt. at 35℃. The polysaccharide preparation purified by ion exchange column chromatography and gel filtration seems to be homogeneous by ultracentrifugal analysis and polyacrylamid gel electrophoresis. Its sedimentation coefficient, 6.28 S. Intrinsic viscosity, 0.46 dl/g. Partial specific volume, 0.67ml/g. Molecular weight, 275,000 and rotation αD1(?)+18. This polysaccharide is composed of D-mannose and D-galactose with molar ratio of 2:1. Pyruvic acid may be also present. Its IR spectrum is similar to that of other polysaccharides.
