长叶车前花叶病毒(HRVsh)外壳蛋白赖氨酸残基经三硝基苯磺酸修饰后的空间结构变化

【摘要】 用紫外光差光谱和圆二色性光谱研究了长叶车前花叶病毒HRVsh在尿素作用下以及经三硝基苯磺酸(TNBS)修饰后外壳蛋白亚基空间结构的变化。TNBS修饰一个赖氨酸残基后的HRVsh外壳蛋白和HRVsh完整颗粒的a-螺旋变化很小。空间结构的变化是局部的。讨论了空间结构变化与生物活力的关系。

【Abstract】 A lysine residue of HRVsh could be modified by trintrophenylsulfonic acid (TNBS) at pH 8.5 in the absence of urea.After modification,the TNP-HRVsh lost its infectivity over ninety percent,brt the virus particle still kept its intact form.The CD spectra in the ultraviolet region suggested that the α-helix of HRVsh was not affected by the modification.Whereas the dif-ference spectra of TNP-coat protein ofHRVsh showed that there are a peak at 230 nm and two shoulders at 260 nm and 290 nm respectively.The TNP-HRVsh shows a difference spectra peak at 260 nm and two shoulders at 230 nm and 287 nm.These results indicated that the modification of a lysine residue of HRVsh with TNBS caused some local alteration of tertiary structure of HRVsh which led to the loss of its in-fectivity.
