大鼠下丘脑弓状核区在唇针镇痛中的作用
ROLE OF HYPOTHALAMIC ARCUATE REGION IN LIP-ACUPUNCTURE ANALGESIA IN RATS
【摘要】 下丘脑弓状核区和脑内参与痛觉调制的结构有复杂的纤维联系,脑内β-脂肪激素-β-内啡肽-ACTH 神经元系统的胞体亦主要集中于下丘脑弓状核区。本实验用电解损毁、H 刀游离和电刺激大白鼠弓状核区的方法,研究该区在痛觉调制和唇针镇痛中的作用。(1)电解损毁或 H 刀游离弓状核区后基础痛阈未见明显改变,但唇针镇痛效应均明显降低。(2)单纯电刺激弓状核区能明显升高基础痛阈;唇针和电刺激弓状核区同时进行时的镇痛效应比单纯唇针时的镇痛效应有明显提高。根据这些结果,讨论了下丘脑弓状核区在大白鼠唇针镇痛中的作用。
【Abstract】 There are a lot of evidences that the hypothalamic arcuate region(HAR)has a close connection with many structures in tho brain which participate in painmodulation.Furthermore,immunocytochemical studies indicate that the cellbodies of the β-lipotropin/β-endorphin/ACTH neuronal systemis is also locatedmainly in the HAR.In this paper,by means of electrolytically induced lesions,surgical isolation and electrical stimulation,the role of the HAR in painmodulation and lip-acupuncture analgesia was studied in rats.The pain thresholds were measured with tail-stimulation/vocalization test.Electro-acupuncture was applied to the“Renzhong”and“Chengjiang”points.Analgesic effect was expressed as a percentage of increase in the pain thresholdafter lip-acupuncture or HAR stimulation.The results were summarized asfollows:1.Effect of HAR lesion:Before and after HAR lesion the baseline pain thres-holds were similar.On the 3rd and 7th days after the lesion the analgesic effectof lip-acupuncture decreased significantly(P<0.01)from the prelesion value259±51 % to 70±28 % and 61±25 % respectively.2.Effect of HAR isolation:Before and after HAR isolation with Halászknife,no marked change was observed in the baseline pain thresholds.However,on the 3rd and 5th days after isolation,the analgesic effect of lip-acupuncturewas also reduced significantly (p<0.05)from the preisolation value 204±39%to 69±29% and 64±24% respectively.3.Effect of HAR stimulation:HAR stimulation not only had marked anal-gesic effect(154±40%, p<0.01),but also enhanced the analgesic effect of lip-acupuncture significantly(from 141±43% to 251±58%, p<0.05).The above results demonstrate that the HAR plays an important role in lip-acupuncture analgesia.
