抗乙型肝炎表面抗原免疫RNA及其mRNA的分离和活性鉴定

【摘要】 本文应用巨噬细胞电泳试验(MEM)和白细胞粘附抑制试验(LAI),证明用热酚法提取的乙型肝炎表面抗原-免疫核糖核酸(HBsAg-iRNA),具有传递细胞免疫反应性的活性。此外,从HB8Ag-iRNA中,用寡脱氧胸腺核苷酸[oligo(dT)]纤维素分离其中的信息核糖核酸(mRNA)。在麦胚无细胞蛋白质合成体系中,证明此种mRNA具有翻译蛋白质的模板作用。应用免疫沉淀法和聚丙烯胺凝胶电泳法提示了乙型肝炎表面抗原-信息核糖核酸(HBsAg-mRNA)所翻译的初生蛋白质或多肽中,含有可与乙型肝炎表面抗原(HBsAg)形成抗体复合物沉淀反应的抗HBsAg的抗体或它的有效肽链。

【Abstract】 It was shown that anti-HBsAg immune RNA isolated by hot phenol method could transfer cellular immune response in the maorophage eleotrophoretio mobility (MEM) and leucocyte adherence inhibition (LAI) tests. Besides, the mRNA was isolated by oligo (dT)-cellulose chromatography and proved to have the activity of protein translation in the wheat germ cell-free protein synthesizing system, the incorporation of 3H-leucine into the translation product of the mRNA being 2-times higher than that of the control without mRNA, By applying the immunopreoi-pitation and polyacrylamide gel electrophoresis, it was indicated that the nascent proteins or peptides translated by HBsAg-mRNA contained the anti-HBsAg antibody or its effective fragments.
