高效液相色谱对酶法合成头孢力新过程的解析
HPLC DETERMINATION FOR PROCESS CONTROL OF CEPHALEXIN PRODUCTION BY ENZYME INDUCTION
【摘要】 酶法合成头孢力新反应过程有四种组分(苯甘氨酸、苯甘氨酸甲酯、7-氨基脱乙酰氧基头孢烷酸和头孢力新),应用日立2632阴离子交换柱和0.05M NaCl 溶于6.25×10-3M NH4Cl作淋洗液,在不到一小时内得到良好的分离。本文报道用高效液相色谱方法对头孢力新反应过程的四种组分进行快速准确的定量。
【Abstract】 The four reaction components(phenylglycine,phenylglycine methyl ester,7-amino-deacetoxy cephalosporanic acid and cephalexin)of synthetic cephalexin obt-ained by enzyme induction were nicely separated within 1 hour,using the Hitachi2632 anionic resin stationary phase and buffered(0.05M NaGl,6.25×10-3M NH4Cl,pH5.5)eluant.The quantitation outlined in this paper allows for the rapid andaccurate determination of cephalexin for process control.
