胸腺激素受体的研究
STUDIES ON THE THYMIC HORMONE RECEPTOR
【摘要】 胸腺激素可促进T淋巴细胞的成熟,因此在细胞免疫功能中起着十分重要的作用,本文报道了以125Ⅰ-胸腺激素T1的放射受体测定法研究猪胸腺激素T1受体的初步结果。结果指出胸腺淋巴细胞的表面有胸腺激素受体,在pH6.0,37℃条件下,激素与淋巴细胞孵育60~90分钟,特异性结合可达最高。向结合系统中加入胰岛素、皮质激素或甲状腺素,不影响胸腺激素T1与受体的结合,证明这种结合是专一的。但在胸腺激素各个组分间,这种专一性并不是很严格,例如胸腺激素E1的性质与T1有很大差别,但可以和T1竞争受体。E1和T1可以竞争同一受体是否暗示它们作用于同一T 细胞亚群,有待证实。胸腺激素与受体的结合能力与它的生物活力是相关的,T1的活力随胰蛋白酶的消化而逐渐丧失,相应的T1与受体结合的能力亦逐渐降低至零。所以用放射受体的方法测定的胸腺激素是代表有生物活力的部分,它的测定范围一般在10~1000毫微克之间。
【Abstract】 Thymic hormones modulating the maturation of T lymphocytes play veryimportant roles in cell mediated immunity.Preliminary results of the studies on the porcine thymic hormone T1 receptorwith radioreceptorassay of 125I-thymic hormone T1 are reported.There are thymic hormone receptors located on the surface of the T lymphooytes,at pH6.0 and 37℃,a maximal specific binding of hormone to lymphocytes can beattained in 60~90 minutes of incubation.With the addition of either insulin,cortison or thyroxin to the assay system,no effect on the binding of thymic hormoneT1 to receptors could be detected.This shows that the binding is specific.Thymic hormone F1 another component of the thymic hormone family,differsfrom T1 in some biochemical properties,but competes with the same receptor;whetherthis fact implies that both affect the same subclasses of T lymphocyte remains to besolved.The binding capacity of thymic hormone T1 to its receptor is parallel to theactivity of the hormone.T1 is degraded gradually by trypsin digestion and simultane-ously loses its capacity to combine with the corresponding receptor.Thus the amountof thymic hormone detected by radioreceptorassay may be considered to represent oneparameter of its biological activity.The measurable range of the assay is between10to 1000ng.
