二亚硝基哌嗪所致大鼠鼻咽上皮非典型细胞动力学变化及人鼻咽癌的细胞动力学研究

【摘要】 本文将有助于了解鼻咽癌的发病机制,并可为临床提供有关的细胞动力学参数。采用阻断动力学结合放射自显影的方法,用长春新硷作丝裂阻断剂,确定其最适剂量(0.83毫克/公斤)及作用持续时间,并用 Ehrlich 腹水瘤验证了方法的可靠性。用二亚硝基哌嗪给大鼠背部皮下长期注射后,鼻咽鳞状上皮的标记指数升高、S 期延长并有增殖区室扩大的现象。用体外脉冲标记法对人鼻咽癌组织的细胞动力学特征作了初步的研究。

【Abstract】 To study the cytokinetics of nasopharyngeal epithelium of rats treated by S.C.injection of N,N’-dinitrosopiperazine(DNP)may be helpful in elucidating themechanism of carcinogenesis.The stathmokinetic and autoradiographic techniqueswere used in animal experiments.Vincristine was selected as the metaphase arrestagent,its optimal dose was 0.83mg/kg b.w.and the arresting effect could last 10 to12hrs.The method was proved to be reliable by Ehrlich ascites tumor because thecytokinetic data obtained from Ehrlich ascites tumor agreed with those reported byclassical method.In normal rats,labeling indexes of the squamous epithelia located atthe lateral and ventral side of nasopharynx were 13.1% and 9.3%,the time intervalsof S phase(Ts)were 6.7 hr and 7.1 hr.respectively.After giving S.C.injection ofDNP 15mg/kg b.w.twice weekly over 179 days,in addition to the increase of LI(18.9% and 20.4%),an expansion of proliferative compartment and a prolongation ofS phase(9 and 10.3 hr.)were induced.It was interesting that these changes were de-monstrated in morphologically normal mucosal epithelium of nasopharynx and theymight represent an essential step in carcinogenesis.The method of in vitro incorporation of ~3H-TdR was used for human biopsyspecimens.The LIs among the squamous carcinomas grade Ⅲ of nasopharynx variedwidely(12.4—33.1%).A shorter generation time and a higher cell loss were foundin the carcinoma with a higher LI.It is suggested that cytokinetic parameters shouldbe studied individually and could serve as a meaningful indication for improving cli-nical therapy.
