苦豆子胶多糖化学结构的测定

【摘要】 <正> 苦豆子胶(Sophora alopecuroides Linn)属豆科槐属多年生的灌木,在我国西部几省分布较广。其种子内胚乳含多糖胶,迄今尚未加以研究和应用,为了开发和合理利用这种丰富资源,我们已开始对苦豆子胶进行各方面的研究。本文介绍对苦豆子胶多糖的化学结构进行测定的初步结果。实验部分

【Abstract】 The polysaccharide isolated from Sophora alepecuroides Linn was hydrolyzed by 2N trifluoroacetic acid, reduced by sodium borohydride to individual alditols and finally converted to the corresponding acetates of various glycosyl residues by acetic anhydride with sodium acetate as the catalyst. GC analysis of the acetates both by the packed column of ECNSS-M and the capillary column of OV-225 show that the molecular ratio of tile glycosyl residues in the polysaccharide is as follows: L-Rha: D-Rib: L-Ara: D-Xyl: D-Man: D-Gal: D-Glu=0.20: 0.03: 0.05: 0.02: 1.83: 1.00: 0.03 The polysaccharide was methylated by using sodium dimethyl sulfinyl anion and methyl iodide. The Feralkylated product was then hydrolyzed and acetylated to get the individual acetates of methylated glycosyl residues. Based on the retention values of the acetates in GC, the ratio of peak areas and the GC-MS and NMR analysis the most probable structure of the polysaccharide is assigned as follows;
