脑啡肽抗血清的制备

【摘要】 为了应用放射免疫测定法和免疫组织化学方法进行脑啡肽的研究,我们制备了抗亮氨酸脑啡肽血清(ALS)和抗甲硫氨酸脑啡肽(AMS)。主要是分别合成两种脑啡肽免疫原,用大白兔进行免疫,经一次强化后分离和制备抗血清,所得抗血清具有满意的灵敏度和精密度。其最小检出量ALS为25pg,AMS为60pg。并具有较好特异性;LEK对MEK的交叉反应率为0.44%,MEK对LEK为2.8%。用效点系统评分法,其结果ALS为30分,AMS为27分,均属较优质的抗血清,能满足于脑啡肽的放射免疫测定要求。

【Abstract】 In order to pursue enkephalin studies by means of radioimmunoassay and immunohistochemical methode, We prepared anti-leu-enkephalin serum (ALS) and anti-met-enkephalin (AMS).The procedure was as follows: Rabbits were immunized with respective enkephalin immunogen of home and foreign make. The specific antiserum was thus obtained. The antiserum possessed satisfactory sensitivity, specificity and repla-ceability for RIA. In the competitive inhibition curves the minimal detactable limit of ALS was 25 pg and that of AMS 60 pg.In the cross test reactions with seven kinds of peptide hormones and morphine, no significant interference was found. The cross reaction rate of LEK vs AMD was 0.44%, and that of AMS vs LEK 2.8%. According to Cerceo’s method of power-point system, both ALS and AMS were reliable antiserums in our experiments.
