百合花丝组织培养及其细胞学观察
TISSUE CULTURE OF LILY FILAMENT AND ITS CYTOLOGICAL OBSERVATIONS
【摘要】 取兰州地区栽培的百合[Lilium davidii var.Willmottiae (Wilson) Roffill]花丝,放入改良MS固体培养基中,在25°±1℃的弱光下培养,20天内花丝不断伸长变粗,末端显著膨大,30天后产生了直径为2mm大小的愈伤组织。转移到分化培养基上,20天后逐渐分化出小鳞茎,40天后形成再生植株。在分化过程中用秋水仙素溶液处理幼小植株,发现经处理的植株不仅具有四倍体植株的外形特征,并且大部分很尖细胞的染色体由原来的24条(2n)变为48条(4n),表明已可能诱发产生了四倍体植株。对愈伤组织进行细胞学观察,发现细胞核的形状、大小及染色体的数目有很大变化,并观察到薄壁细胞间的染色质穿壁运动。
【Abstract】 Anthers with the filament of lily [Lilium davidii var. Willmottiae (Wilson) Rof- fill] were planted on modified MS medium. Filament cultures were incubated at 25°± 1℃, and illuminated with a fluorescent light of about 800--1200 Lux. Cultured fila- ments were initially short in length, but gradually became thicker and elongater after 20 days. After 30 days, the calli (about 2 mm in size) were produced. The calli were then transferred to the differentiation medium. After 20 days, the bulblets were de- veloped from the calli, and the regeneration plants were formed after 40 days For tetraploid induction, plantlets were treated by aqueous solution of colchicine. A preliminary ploidy analysis of the root tip cells of these treated plants indicated that it was predominantly tetraploid (4n=48). Of 56 mitotic figures examined, 19 were tetraploid, 10 were diploid and 27 were aneuploid. The intercellular migrating chromatin substance appeared in the calli were also observed.
