利用EB病毒特异性DNA酶检测鼻咽癌的研究

【摘要】 <正>利用病人血清的免疫学及生物化学测定方法探测体内恶性肿瘤,是癌瘤早期发现、早期诊断所的重要方法,此法在检查中不影响病人健康,利于大规模人群普查。近年来,文献资料一致指出Epstein—Barr病毒(EBV)与鼻咽癌(NPC)密切相关,尽管其病因学的地位尚未确定,但NPC病人血清中对抗EBV的某些抗原,如早期抗原(Early antigen,EA)、病毒壳抗原(Virus capsid antigen,VCA)的抗体(IgA或IgG型)的出现及其滴度高低可作为鼻癌咽早期发现、早期诊断、预后推测等的重要参考。

【Abstract】 It is an important approach to develop new methods for early detection and diagnosis of cancers by the use of immunological and biochemical assays of patients’ sera, in recent years a lot of literature has pointed out unanimously that there is a close relationship between Epstein-Barr virus ( EBY ) and na sopha ry ngeal carcinoma (NPC). In 1980, an EBV spoeific DXAse was found by Cheng Y C in Raji cells super-infected by EBV, produced by P3HR-1 cells. Recently, we have shown that this EBV, specific DNVse can also bo Indren d by the activation o f croton oil and n-butyric acid in EBV ge nome- containing cell lines: Raji and P3HR-1 cells.In order to test the availability of this enzyme analysis for the detection of NPC, a comparison of the activities of DXYse from the Raji or P3HR-1 cells after the addition of sera from normal person: or NPC patients, was made. It was shown that the addition of NPC sera decreased the Raji DXAse activities to 45 - 48% of the values for normal sera and the differences were very significant ( P<0.001 ) . The false negative ( D P M values higher t h an average norina1 control) rates was shown to be about 5% . Some parallelism between the anti DNAse activities and anti VCA-IgA titers of patients’ sera was also found, but this will be discussed elsewhere.In the experiments with P3HR-1 cells, sera from NPC patients decreased the DNAse activities to 52.7-56.4% of normal value (P<0.05). With the improvement and perfection of this method we expect that it can be used clinically to detect and diagnose XPC. to conduct perspective investigation in NPC high risk population and also to monition the therapeutic responses and recurrences of NPC after radiation and chemotherapy.
