柑桔胚培养技术的研究——Ⅱ、多胚性品种未熟幼胚的早期离体培养

【摘要】 本文是叙述多胚性柑桔品种未熟幼胚的早期培养技术,有可能为提高育种效率提供一条更有效的途径。使用的是椪柑( Citrus reticulata Blanco)和香水橙 ×枳( citrus sinensis × Poncirus trifoliata)的幼小种子。开花后105天的椪柑种子接种在附加IAA(1ppm)、BA(0.2ppm)、CH(400ppm)的MS培养基上,其中的幼胚能继续“胚性生长”并且增大,同时还形成了大量的胚状体。当这些增大了的胚及胚状体转移到含有NAA( 0.75 ppm)的MS培养基上后,能分化根、茎、叶,并发育成正常的植株。对授粉后75天的香水橙×枳的幼胚也作了诱导,并获得初步成功,它们的部分原胚形成了植株。

【Abstract】 The technique of embryo culture in vitro with immature seeds of ’Ponkan’ ( Citrus reticulata ) and ’Xiang-shui-chen’ x trifoliate orange ( C. sinensis x Poncirus trifoliata ) is described. It may provide a more effective means in the breeding of polyembryonic varieties.Immature seeds of ’Ponkan’ 105 days after flowering were placed on the MS medium supole-mented with IAA ( 1 ppm ) , CH (400 ppm) and BA (0.2 ppm). The growth of the small embryos in the immature seeds continued and the small embryos enlarged. At the same time, a large number of embryoids were produced in these immature seeds. When the enlarged embryos and embryoids were transplanted onto MS medium containing NAA ( 0.75 ppm ) , they formed rqots, shoots and leaves and developed into normal plants.Embryos of immature seeds from the cross ’Xian-shui-chen’ x trifolate orange 75 days after polli-nation were cultured and several seedlings were also obtained.
