伞形科中药的研究——Ⅲ、长前胡的新香豆素

【摘要】 <正> 长前胡(Peucedanum turgeniifolium Wolff)为伞形科前胡属植物,药用全草,能宣散风热,祛痰镇咳,下气;治感冒,咳嗽,痰稠,头痛及胸闷等症。其化学成分未见报道,为此我们对全草的化学成分进行了研究。 此次研究材料系四川成都中药材采购供应站提供。提取分离方法如下:将全草磨粉,用甲醇抽提,得绿褐色总抽出物约为6.5%,活性炭脱色至浅黄色,浓缩至小体积放置,析出大量白色细针晶,经鉴定为甘露醇,得率约1.2%。母液经中性氧化铝(层析用,

【Abstract】 The total plant of Peucedanum turgeniifolium Wolff were extracted with methanol and the extract was chromatographed over alumina. Five dihydroseselin type coumarins were obtained. The structure of three new coumarins of them, respectively named turgeniifolin A ( Ⅰ ) , turgeniifolin B (Ⅱ) , turgeniifoselin C (Ⅱ) , were determined by chemical and spectroscopic data (UV, IR, MS, PMR and CMR) .Turgeniifolin A (Ⅰ), C18H18O6, mp. 198-201℃, [α] D18±0°(CHC13) . The structure of (Ⅰ) was elucidated as 3 ’ -angeloyloxy- 4 ’ -carbonyl- 3 ’, 4’-dihydroseselin. CMR data of ( Ⅰ ) at δ (ppm) :160.0 (C2) , 113.9 (C3) ,143.3 (C4) ,135.1 (C5) ,114.9 (C6) ,159.6 (C7) , 114.0 (C8) , 154.7 (C9) , 113.1 (C10) , 82.4 (C2’) , 76.5 (C3’) , 184.5 (C4’), 26.3 and 19.7 (C5’and C6’) , 166.1 (C1") , 126.9 (C2") , 139.9 (C3") , 20.5 and 16.0 (C4"and C5") .Turgeniifolin B (Ⅱ) , C18H22O6, mp. 160-162℃,[α]D18±O° (CHC13). The structure of (Ⅱ) was elucidated as 3 ’ -isovaleryloxy- 4 ’ -hydroxy-dihyd-roseselin. CMR data of (Ⅱ) at δ (ppm) :161.1 (C2), 113.5 (C3) , 144.1 (C4) , 128.6 (C5) , 114.7 (C6) , 155.9 (C7) , 113.5 (C8) , 154.6(C9), 110.4 (C10) , 77.2 (C2’) , 73.9 (C3’) , 62.7 (C4’) ,24.0 and 23.9 (C5’ and C6’) 172.0 (C1"), 43.2 (C2") 24.30 (C3") , 22.3 and 25.6 (C4"and C5") . Turgeniifolin C (Ⅲ) , C19H20O6 mp. 201-203℃[α]D18±0° (CHC13) .The structure of (Ⅲ) was elucidated as 3 ’-senecioyloxy- 4 ’-hydroxy-dihyd-roseselin. CMR data of (Ⅲ) at δ (ppm) :161.0 (C2) ,113.5 (C3) , 144.1(C4) , 128.6 (C5) , 114.8 (C6) , 156.1 (C7) , 113.5 (C8), 154.6(C9) ,110.3 (C10) , 77.1 (C2’) , 72.9 (C3’) , 62.8 (C4’) , 24.1 and 23.8 (C5’and C6’) , 165.2(C1"),115.4(C2"), 158.9(C3"),20.4 and 27.4 (C4"andC5") .The other two coumarins, ( ± ) -cis-khellactone (Ⅵ) and (±) -trans-khellactone (Ⅴ) , are found for the first time in nature.(±) -cis-khellactone (Ⅳ) , C14H14O5, mp. 153-155℃, [α]D23±0° (CHC13) .CMR data of (Ⅳ) at (ppm) :162.5 (C2) , 111.9 (C3) , 145.1 (C4) , 129.0 (C5) , 115.2 (C6) , 156.9 (C7) , 112.8 (C8) , 152.4(C9), 111.8 (C10) , 79.4 (C2’) , 71.7 (C3’) , 60.9 (C4’) , 25.8 and 21.2 (C5’ and C6’) .(±) -trans-khellactone (Ⅴ) , C14H14O5, mp. 186-188℃ [α]D23±0°(CHC13) . CMR data of (Ⅴ) at (ppm) :161.1 (C2) , 111.9 (C3) , 145.6(C4) , 129.0 (C5) , 115.4 (C6) , 155.1 (C7) , 112.9 (C8), 152.3(C9) ,112.2 (C10) , 79.8 (C2’) , 74.9 (C3’) , 66.1 (C4’), 25.2 and 21.5 (C5’and C6’) .
