毛霉酶促磷酸化反应条件的研究
Studies on the conditions for reaction of enzymatic Phosphorylation of mucor aromaticus povah
【摘要】 本文较详细的研究了毛霉酶促磷酸化的反应条件,并讨论了反应体系中各种因素对其酶促磷酸化的影响。在试验中发现高浓度葡萄糖能推迟ATP累积高峰出现的时间;高浓度磷酸盐能促进磷酸化反应;葡萄糖、磷酸盐和反应时三者的配合关系对ATP克分子转化率影响较大。最后用正交试验确定了酶促反应的较佳方案。
【Abstract】 In the reaction system of enzymatic phosphorylation of M. aromaticus, the time of the geak accumulation of ATP could be postponed by the high concentration of glucose; high concentration of phosphate could promote the enzymatic phosphorylation. The coordination of glucose, phosphate and reaction time has a decisive effect on gramme molecule change effiency of ATP. One gramme of fresh mycelia changes 75 M AMP into ATP under appropriate laboratory. The appropriate conditions of reaction were: glucose 250, K2HPO4 350, AMP 30, Mgcl2 30 μMol/ml, PH7.5, methybenzene 5%, fresh mycleia 40% (w/v), 35℃, 3hrs.
