影响小鼠混合淋巴细胞反应的实验条件分析

【摘要】 <正> 混合淋巴细胞反应(MLR)可定量反映个体间组织相容性抗原遗传型的差异程度,亦可检测淋巴细胞的反应能力。目前已被广泛用于组织配型,免疫耐受和免疫活性细胞及其亚群之间的相互关系等方面的研究。由于小鼠的遗传型可经基因重组的方式加以改造,培育出具有一定遗传型的近交系小鼠,因而小鼠MLR常用为研究移植免疫的一种实验方法。MLR的测定通常以氚标记的胸腺嘧啶核苷(~3H-TdR)掺入量作为指标。

【Abstract】 The results of an investigation on some of the experimental factors that operated during the mouse mixed lymphocyte culture (MLC) were reported. The following experimental conditions were considered to be optimal for maximal reaction. (1) Mouse lymph node cells were suspended at concentration of 2×106 cells/ml in RPMI-1640 medium supplemented with 15% inactivated human AB serum and antibiotics. (2) Responding and stimulating cells were mixed equally in total of 0.4 ml in 10×75 mm round-bottomed tube and incubated for 4 days at 37℃ in CO2-incubator. (3) Each culture was pulse labelled with 0.5μCi/25μl 3H-thymidine (sp. act. 5Ci/mM) for 16 hours before harvest. This test system was proved to be reliable to discriminate clearly the response of allogeneic lymphocytes between two strains of inbred mice. The response in MLO system might be inhibited by contaminating red cells and platelets in cell suspensions. Human serum and 2-mercaptoethanol could improve the response in MLO and promote a nonspecific stimulation in control cultures.
