碳水化合物的合成研究 Ⅰ.D-果糖-1-磷酸酯及D-果糖的酶促合成

【摘要】 本文报道了用化学与酶促相结合的方法,由二羟基丙酮磷酸酯与D-甘油醛在兔肌醛缩酶的存在下进行酶促不对称合成,一步缩合得到D-果糖-1-磷酸酯,经酸水解后得到结晶D-果糖。用同样的方法,以DL-甘油醛代替D-甘油醛则得到D-果糖-1-磷酸酯和L-山梨糖-1-磷酸酯的混合物。用钙盐结晶的方法分离,得到D-果糖-氯化钙结晶,除去氯化钙后得到单一的结晶D-果糖。母液在Dowex-1 B4O7-型树脂进行梯度淋洗得到L-山梨糖结晶。
【Abstract】 The present report deals with the asymmetric preparation of D-fructose-1-phosphate and D-fructose. The phosphate was prepared through one step condensation of dihydroxyacetone-phosphate and D-glyceraldehyde in the presence of aldolase. Crystalline D-fructose was obtained after acid hydrolysis of the phosphate. In a similar way, a mixture of D-fructoso-1-phosphate and L-sorbose-1-phosphate was prepared by using racemic glyceraldehyde instead of D-glyceraldehyde. The mixture of the sugar phosphate was hydrolyzed by dilute acid and the hydrolysis products were then separated by the addition of calcium chloride. The D-fructose was formed as crystalline fructose·CaCl2, while the sorbose remained in the mother liquior and was recovered by using chromatographic fractionation. By such method, pure crystals of D-fructose and L-sorbose were obtained. The crystals of D-fructose and L-sorbose were identified by thin layer chromatography, optical rotation, gas chromatography and IR spectroscopy.
