【Abstract】 Adenosine deaminase deletion is known to be associated with the condition of severe combined immunodeficiency. A spectrophotometric method for measurement of the enzyme activity of adenosine deaminase in blood cell is described. The method is based on Kalckar’s principle and established according to our own condition. Comparing to more sophisticated methods, we find that our procedure is simple, reproducible and reliable and requiring no special equipment. The blood volume needed for sampling is small. Some of the shortcomings of the method are discussed. Using this method adenosine deaminase activity in normal human red blood cells was found to be 2.7±1.0 units, and in lymphocytes, 32±14 units. The results were similar to those recorded in foreign literatures. The enzyme activity in two children diagnosed as cellular immunodeficiency were found to be lower than normal in the red blood cells, while another case of humoral immunodeflciency child gave normal enzyme activity value. For determination of the correlation between adenosine deaminase activity and immunodeficiency disorders, further works have to be done.
