条斑紫菜Porphyra yezoensis Ueda营养细胞的分离和培养

【摘要】 <正> 条斑紫菜营养细胞在一定条件下(主要是温度)可转化成单孢子,经放散、附着,再发育成新的藻体。但单个营养细胞在未形成单孢子以前,能否发育成新藻体尚未见报导。已知海带(Laminaria japonica)、凋毛藻(Griffithsia pacifica)、尾丝藻(Uronema gigae)、石莼(Ulvapertusa Kjellm),浒苔(Enteromrpha intesinalis(L.)Grer.)和若干种高等植物的营养细胞能在适当的条件下发育成新的植物体。条斑紫菜的营养细胞有无这样的发育全能性,对此,我们从1979年3月开始,反复进行了四次实验。结果表明,这种紫菜营养细胞的发育全能性是存在的。

【Abstract】 1,Individual vegetative cells of Porphyra yezoensis were obtained by mechanical smashing and filtering. After isolation, cultioation, the cell wall of the isolated cells began to become thickened and the cells each began to divide and developed into a new frond. The vegetative cells of Porphyra yezoensis have been observed to be totipotent in development. The furvival rate of the isolated cells was about 33%.2, The isolated cells could develop into new fronds under4 °-22℃, but the optimal temperature was found to be 12°-13℃.3 , Special supplementary culture and continuous illumination seemed to be able to quicken the developing process.
