四种噻二唑类化合物对妊娠大白鼠致畸活性的比较研究

【摘要】 本文主要讨论了四种噻二唑类化合物对妊娠大白鼠的胚胎毒作用和胎鼠毒作用。根据我们的实验结果,可以认为: 1.四种噻二唑类化合物对妊娠大白鼠的胚胎毒性和胎鼠毒性,在强度上表现为下列关系: Bis-A-TDA>A-TDA>Bis-HM-A-TDA>Bis-A-S-TDA 2.四种噻二唑类化合物在活性上的差异与它们化学结构上的某些活性基团密切相关。

【Abstract】 The present experiment was conducted with sexually mature, virgin female rats. The body weight of the female rats was 155~250g and that of the males was 280~320g. The females and males were mated over-night, ♀: ♂ ratio of rats was 3:1. The female rats were considered O-day of gestation when the sperms were found in the vagina. Then the pregnant rats were divided into eight experimental and one control (1% solution of methyl cellulose) groups.The drugs was administered (p. o.) on the 10th day of gestation with the dosage of Bis-A-TDA 5 and 7.5mg/kg; that of A-T-DA and Bis-HM-A-TDA 5 and 20mg/kg and Bis-A-S-TDA 5 and 120mg/kg respectively.All pregnant rats were killed on the 20th day of gestation, the number of corpora lutea, implantation, resorption sites and live fetuses were registered. The living fetuses were removed, weighed, and inspected for gross abnormalities and sexes; the internal and skeletal malformations of the live fetuses were examined by method of Wilson.Under present experimental conditions, the teratogenic activity of four thiadiazole compounds was of the following sequence:Bis-A-TDA>A-TDA>Bis-HM-A-TDA>Bis-A-S-TDA;This may be due to the presence of different active groups in the four compounds.
